IDRA-21 functions as a positive allosteric modulator of AMPA glutamate receptors (ampakine), representing a benzothiadiazine derivative that acts as a chiral molecule with (+)-IDRA-21 being the active form. The compound partially allosterically modulates AMPA receptor desensitization, enhances long-term potentiation (LTP) between synapses in the brain, inhibits AMPA receptor desensitization, and increases excitatory synaptic strength, with blood-brain barrier permeability enabling CNS penetration. IDRA-21 shows nootropic effects in animal studies, significantly improving learning and memory, demonstrating approximately 10–30 times greater potency than aniracetam in reversing cognitive deficits induced by alprazolam or scopolamine (with some studies reporting 1030 times greater potency), and producing sustained effects lasting for up to 48 hours after a single dose.
