Research Applications & Storage Protocols

Lipophilic Glutathione Prodrug & Antioxidant Activity Profile

N-Acetylcysteine ethyl ester (NACET, NAC-Et) is an antioxidant which acts as a lipophilic prodrug for N-acetylcysteine and subsequently glutathione, with improved bioavailability over N-acetylcysteine itself. The additional ethyl ester group enhances cellular uptake and bioavailability compared to N-acetylcysteine (NAC). NACET is best-known for its ability to improve intracellular glutathione levels and has fascinating antioxidant properties to help support healthy ageing. It increases glutathione levels in rat liver, kidney, heart, testis, and brain when administered at a dose of 50 mg/kg twice per day for two weeks. NACET has been researched in animal studies as a potential treatment for diabetic retinopathy, where its antioxidant effects are potent in the eyes, protecting retinal cells against oxidative damage. It has also been sold as a “dietary supplement” in combination with glycine under the name GlyNAC-Et as a supposedly improved version of the older glycine-NAC combination GlyNAC, which has been claimed to show efficacy in counteracting symptoms associated with aging as well as HIV infection, diabetes and nerve damage for in vitro/inivo modeling applications. NACET can be used in sports nutrition supplements to reduce oxidative stress after exercise and aid in body recovery.
