Semax is a medication used in Eastern Europe for treatment of brain trauma and predominantly for its claimed nootropic, neuroprotective, and neurorestorative effects. Chemically, Semax is a heptapeptide and synthetic analogue of a fragment of adrenocorticotropic hormone (ACTH), ACTH(4-10), with amino acid sequence Met-Glu-His-Phe-Pro-Gly-Pro (MEHFPGP). The heptapeptide Semax is completely devoid of any hormonal activity but enhances learning and memory abilities. The most important function of Semax is stimulating BDNF (brain-derived neurotrophic factor) synthesis, a peptide secreted by neurons necessary for retinal function (greater contrast and sharper vision), cholinergic and dopaminergic neuron function, long-term memory formation, and development of new nerve connections. Semax has been found to stimulate memory and attention in rodents and humans after intranasal application, augment effects of psychostimulants on central dopamine release, improve selective attention, and modulate brain development.
