Delta Sleep-Inducing Peptide (DSIP) was isolated from rabbit cerebral venous blood by Schoenenberger-Monnier group from Basel in 1977 and initially regarded as a candidate sleep-promoting factor. DSIP was shown to be a nonapeptide of molecular weight 849 and to induce mainly delta-sleep (deep slow-wave sleep) in rabbits, rats, mice, and humans, whereas in cats, the effect on REM sleep was more pronounced. In addition to sleep, the peptide affects electrophysiological activity, neurotransmitter levels in the brain, circadian and locomotor patterns, hormonal levels, and psychological performance. Experimental results suggested a modulation or ‘programming’ interaction of DSIP with endogenous opioid-peptidergic systems and exogenous intracerebrally or systemically administered morphine and amphetamine. DSIP induces cerebral MAO-A activity and has a pronounced influence on the circadian rhythms of locomotion and intracerebral neurotransmitter as well as plasma protein and cortisol concentrations. DSIP was also shown to counteract experimentally induced stress situations in animals. An improvement of the psychomotor performance and concentration capacity in humans beside sleep normalization and pronounced effects on withdrawal symptoms including pain states in alcoholics and opiate addicts was discovered. This encouraged a pilot study for action of the peptide in humans suffering from chronic pronounced pain episodes, and DSIP lowered significantly the pain levels of 6 out of 7 patients after intravenous administration, with simultaneous significant reduction of concomitantly occurring depressive states. DSIP-like immunoreactivity was found by RIA and immunohistochemistry in brain and by RIA in peripheral organs of the rat as well as in plasma of several mammals. Plasma DSIP-like immunoreactivity decreased at the transition from wakefulness to sleep, suggesting that DSIP is induced by mechanisms involved in the initiation of sleep. However, the link between DSIP and sleep has never been fully characterized, in part because of the lack of isolation of the DSIP gene, protein, and possible related receptor, making the hypothesis regarding DSIP as a sleep factor extremely poorly documented and still weak. Despite this, DSIP has been examined for therapeutic effects in chronic pain, insomnia, mood improvement, stress regulation, sleep quality, substance withdrawal, and neuroprotection.
